【Pharmacological Effects】 Pharmacodynamics - Enrofloxacin is a broad-spectrum bactericidal agent specially used for fluoroquinolones. It has a good effect on Escherichia coli, Salmonella, Klebsiella, Brucella, Pasteurella, Actinobacillus pleuropneumoniae, erysipelas, Proteus, Serratia marcescens, Corynebacterium pyogenes, Bordetella septica, Staphylococcus aureus, mycoplasma and chlamydia, but it has a weak effect on Pseudomonas aeruginosa and Streptococcus, and a weak effect on anaerobic bacteria. There is obvious post antibiotic effect for sensitive bacteria. The antibacterial mechanism of enrofloxacin is to inhibit bacterial DNA gyrase, interfere with the replication, transcription, repair and recombination of bacterial DNA, and the bacteria can not grow and reproduce normally and die. Tylosin is a macrolide antibiotic. By binding with the 50S subunit of pathogenic microorganism nucleosome, it inhibits the translocation reaction catalyzed by translocation enzyme, hinders the growth of skin chain of sensitive microorganisms, and affects the synthesis of protein, thus showing antibacterial effect. It can kill mycoplasma, gram-positive bacteria and some gram-negative bacteria.
Pharmacokinetics - This product is absorbed rapidly and completely by intramuscular injection, and its bioavailability is 91.9% in pigs and 82% in cows. It is widely distributed in animals and can enter tissues well. Except cerebrospinal fluid, the drug concentration in almost all tissues is higher than that in plasma. The main metabolism of liver is to remove the ethyl group of 7- piperazine ring to produce ciprofloxacin, followed by oxidation and glucuronic acid combination. It is mainly excreted through the kidney (by renal tubular secretion and glomerular filtration), and 15% ~ 50% is excreted from the urine in its original form. The elimination half-life is quite different in different species of animals and different routes of administration. The elimination half-life after intramuscular injection is 5.9 hours for cows, 1.5 ~ 4.5 hours for sheep and 4.6 hours for pigs. After intramuscular injection, tylosin can be quickly absorbed and spread to tissues, organs and body fluids. The maximum plasma concentration can be reached after 30min of injection. It can also pass through the blood-brain barrier, blood-eye barrier, placental barrier and gonad. The effective blood concentration can be maintained for 24 hours after a single injection. The main way of tylosin excretion is with urine and bile (with feces).
【Drug Interactions】
(1) Heavy metal ions such as Ca2+,Mg2+, Fe3+ and AI3+ can chelate with this product, which will affect its absorption.
(2) When it is combined with theophylline and caffeine, the binding rate of plasma protein will decrease, the concentration of theophylline and caffeine in blood will increase abnormally, and even theophylline poisoning symptoms will appear.
(3) This product has the function of inhibiting liver drug enzymes, which can reduce the clearance rate of drugs mainly metabolized in the liver and increase the blood drug concentration.
【Indications】Antibacterial drugs. Used for bacterial diseases and mycoplasma infection of livestock and poultry.
【Dosage and Administration】 Intramuscular injection: one dose, 0.025ml for cattle, sheep and pigs per 1Kg body weight; 0.025~0.05ml for dogs, cats and rabbits. Once or twice a day for 2-3 days.
【Adverse Reactions】(1) Degenerate the cartilage of young animals, affect the development of bones and cause claudication and pain. (2) The reactions of digestive system include vomiting, loss of appetite and diarrhea. (3) Skin reactions include erythema, itching, urticaria and photosensitive reaction. (4) Dogs and cats occasionally have allergic reactions, ataxia and seizures.
【Precautions】(1) It has potential exciting effect on the central system and induces seizures. Dogs with epilepsy should use it with caution. (2) Carnivores and animals with renal dysfunction can occasionally crystallize urine with caution. (3) This product is not suitable for dogs before 8 weeks old. (4) The drug-resistant strains of this product are increasing, so it should not be used for a long time at sub-therapeutic dose.
